ULTRASTRUCTURAL CHANGES OF STROMA OF GANGLIA OF SPLEEN PLEXUS FOLLOWING CHEMICAL DESYMP A TIZA HON

Абстракт

Recent years researches point to considerable advantage of chemical desympatization of animals under experiment in comparison with surgical and

immune ones for solving a number of problems of structural and functional organization of vegetative nervous system. Animals: 36 adult male rats (200 -225 g). Chemical sympathectomy was produced with intraperitoneal injections of 6-oxyhydrodopamine (6-0HDA),100 ~g /100 g body wt, on Days 1, 3, 5 of the experiment. The 6-0HDA was dissolved in sterile saline. Age matched controls received an equivalent volume of saline but without 6-0HDA. The animals were sacrificed by decapitation on Days 1,3,5 after the last injection of6-0HDA was made. The blocks were fixed in 4-oxide osmium, then were placed in araldite and examined with high performance electronic microscope. The most important change of stroma elements of sympathetic ganglia under desympatization was in considerable thickening of their connective tissue and widening of blood vessels. Quick increase of connective tissue volume alongside

with nervous cells ruin resulted in sclerosis of ganglia and decrease of their sizes. Abundant grow of ganglia connective tissue of spleen plexus is due to proliferation of fibroblasts which has been confirmed by the presence of rbrotlasts with active synthetical activity. There .vas observed dilated profiles of cisterns of g'a-Jar endoplasm network, enlarged quantity of connected ribosomes. There were also noticed ce s with abrupt hypertrophic synthetical scca-atus of albumen and well-developed a~e ar Goldji complex containing contracted : s:e'"s and hyperplasmatic vesicular elements _-e.enly filled with granular substance of a.e'age electron density. In such cells there were
revealed small and large secondary lizosomes. It is worth mentioning that alongside with such cells there were noticed fibroblasts with low activity, with poor development of granular cytoplasmic network and a few ribosomes. There were also noticed degenerating cell shapes. Such cells as a rule were placed among hotbeds of loosening on intracellular substance with low electrone activity, fragmentation and disintegration of collagen fibers. Presence of such fibroblasts in ganglia stroma of spleen plexus can be considered as consequence of toxic effect of 6-OHDA and presence of active fibroblasts with hypertrophy of albumin.-synthesized apparatus as a respond to neuron and part of connective elements ruin. In the result of chemical desympatization in ganglia stroma of spleen plexus there are taking place two opposite processes:! strengthening of collagen-formation due to intensification of synthesis;

2. resorbtion of collagen Sclerosis of nerve ganglia watched by us is probably connected with violation of balance between synthesis and resorbtion of collagen with synthetical processes growing. Alongside with sclerosis processes in stroma there were watched leucocytic infiltration at the spots of neurons destruction and also along blood vessels and collagen fibers bundles both about ganglia periphery and at central part, too. At such places with electron-microscope visualization there were revealed lymphocytes, macrophags, granulocytes and plasmatic cells. Summing up, chemical desympatization by 6-OHDA brings about in sympathetic ganglia of spleen plexus not only destruction and elimination of most part of neurons but also a number of changes of stroma elements of ganglia, development of autoimmune inward processes evidence of which was the presence of plasmatic cells.
